Choice of dialysis tubing for DNA purification | amidon7 | 2001-08-03 | Click here to register. |

I would like to purify a ~12kb DNA fragment from a restriction digest, for pronuclear injection of one cell mouse embryos. Which dialysis tubing would you recommend for this purpose? I would prefer one that is compatible with electro-elution of this DNA fragment from an agarose gel slice.
Thanks very much for your help

 | A good place to start is Spectra/Por 4 (when in doubt use Spectra/Pos 4).
While your DNA fragment has a very high molecular weight, it is a linear and not globular molecule. This allows it to escape from tubing with a smaller MWCO than the fragment weight.
Any of the cellulosic membranes would be usable in your electroelution apparatus. If you feel that the MWCO of the Spectra/Por 4 is to low, you can use any of the Spectra/Por 6 or 7 membranes instead. |
