Difference between CE membrane tubing /ultrafiltration? | taiga | 2001-05-16 | Click here to register. |

Hello,
Could you help me to choose the type of membrane (CE membrane tubing or Regenerated cellulose membranes)I think it's CE membrane tubing MWCO 500 D, but have yout got directive to choose width and length? I would like to separate SeO3 = PM 127 from peptides.
For the time of dialysis, how do you do to choose it? I'll try differents time between 1 - 20 min?
Have you got information to prepare membrane befor to put solution into?
To make dialysis, is it correct if I put solution aqueous in tubing and water in reservoir to have separation?
Thank you very much
Armelle

 | The 500 D MWCO tubing would be good if your peptides are all 5,000 molecular weight or larger. The width and length selection is based mostly on your sample sizes. You should generally use the skinniest tubing (to minimize the diffusion time) that doesn't make a sack that's too big to handle easily (the product of the length of the tubing and its cross-sectional area determine it's volume).
Since dialysis is driven only by diffustion, generally between 4 and 48 hours are required for a good equilibrium.
The solution outside the bag should be as similar to the solution inside the bag as possible. Since water can get through the tubing, it also tries to move from high concentration to low concentration. If the concentration of water is higher outside the bag than inside, water will migrate in and may burst the bag. (Pure water is concentrated water. As you add things to water, the concentration of water goes down while the concentration of the new things goes up.) |
