Hello,
I want to purify bacterial DNA after enzymatic lysis of a suspension of bacteria. I want to concentrate DNA who have a lenght about 50 Kb and eliminate proteins and peptides that appear during the lysis. Which membrane could you recommend me.
Thank you

Karine:

I would recommend either a 50,000 MWCO Spectra/Por 6 or a 100,000 MWCO Spectra/Por CE membrane to start. The DNA strand will be large in terms of molecular weight (more than 5 million daltons for 50,000 base-pairs), but it will be stringy and not globular. Since it's stringy it will be more likely to go through a smaller pore than a similarly weighted globular protein. The 50k/100k MWCO should let most of the proteins and peptides out but still retain most of the DNA.

Let me know how it works out.

Thanks for visiting.
